Pathophysiology of Diabetes Mellitus Type 2: Roles of Obesity, Insulin Resistance and -Cell Dysfunction

چکیده

The past two decades have seen an explosive increase in the number of people diagnosed with diabetes mellitus worldwide, particularly type 2 diabetes (T2D), which is found associated with modern lifestyle, abundant nutrient supply, reduced physical activity, and obesity. Actually, between 60 and 90% of cases of T2D now appear to be related to obesity. Numerous studies have shown that insulin resistance precedes the development of hyperglycemia in subjects that eventually develop T2D. However, it is increasingly being realized that T2D only develops in insulin-resistant subjects with the onset of -cell dysfunction. It is therefore important to characterize the mechanisms of insulin resistance and subsequent pancreatic -cell failure associated with obesity in order to better understand the pathophysiology of T2D and develop approaches to prevent T2D. Impact of Magnesium Deficiency on Pancreatic β-Cell Function in Type 2 Diabetic Nigerians

Objective: Pancreatic b-cell dysfunction is described to be present at the diagnosis of type 2 diabetes mellitus (T2DM) and progressively deteriorated with disease duration. However, its progression is variable and potentially influenced by several factors. The Magnesium (Mg) deficiency mediates insulin resistance but reports regarding its role in pancreatic β-cell dysfunction are scarce and co...

Pathophysiology of type 2 diabetes.

Type 2 diabetes mellitus is a heterogeneous syndrome characterized by abnormalities in carbohydrate and fat metabolism. The causes of type 2 diabetes are multi-factorial and include both genetic and environmental elements that affect beta-cell function and tissue (muscle, liver, adipose tissue, pancreas) insulin sensitivity. Although there is considerable debate as to the relative contributions...

INVESTIGATION OF METHYLATION OF TNF-Α GENE PROMOTER IN PATIENTS WITH TYPE 2 DIABETES

Background: Type II diabetes is a chronic inflammatory condition that is associated with a combination of genetic and environmental factors. Tumor necrosis factor alpha or TNF-α as an adipocyte cytokine, which affects the signaling pathway of insulin, can contribute to insulin resistance in type 2 diabetes patients. Considering the importance of epigenetic changes in multifactorial diseases, th...

Pathophysiology of Type 2 Diabetes in Koreans

The pathophysiology of type 2 diabetes is characterized by variable degrees of insulin resistance and impaired insulin secretion. Both genetic and environmental factors serve as etiologic factors. Recent genetic studies have identified at least 83 variants associated with diabetes. A significant number of these loci are thought to be involved in insulin secretion, either through β-cell developm...

Background: Adiponectin is an adipose tissue-derived hormone that low levels of this hormone are associated with obesity, insulin resistance, and type 2 diabetes. The aim of this study was to compare the serum levels of adiponectin in diabetic and non-diabetic obese individuals. Methods: As a cross-sectional study 35 obese individuals with type 2 diabetes mellitus and 35 non-diabetic obese subj...
